摘要
目的了解东莞市不同人群中艾滋病病毒(HIV)各亚型毒株的流行情况和传播规律。方法采集东莞市经蛋白印迹法确认的24名HIV感染者的外周抗凝血,分离单核细胞、提取核酸后用套式聚合酶链反应扩增HIV-1膜蛋白(env)基因C2-V3区的核酸片段,进行序列测定,鉴定病毒亚型。用威斯康星GCG软件进行共享序列、基因离散率的计算和毒株的聚类分析。结果在24份阳性标本中,有13份标本PCR扩增阳性,亚型分析结果表明东莞市存在HIV-1B和CRF01-AE两种亚型。结论HIV-1B和CRF01-AE可能是东莞市艾滋病病毒流行的主要亚型。
Objective To study the prevalence status and transmission patterns of different subtypes of HIV-1 in Dongguan city. Methods Peripheral mononucleated ceils were prepared from 24 HIV carriers confirmed by Western Blotting. The provirus DNA was extracted and amplified by nested-PCR, after which the HIV-1 membrane protein gene (env) C2-V3 region was sequenced and its subtype was analyzed. Analyses of the consensus sequence, gene divergence and clustering of virus strains were conducted with Winsconson C_,CG software. Results Among 24 anti-HIV-1 antibody positive samples, 13 HIV-1 env gene fragments were amplified and analyzed. Subtypes B and E of HIV-1 virus strains were found in Dongguan city. Conelusion Subtype B and E of HIV-1 are probably main epidemiological strains in Dongguan.
