摘要
本试验将新城疫(ND)疫苗与B11001株鹦鹉热衣原体混合,用ND阳性血清中和,每个接种剂量最终含ND疫苗10羽份,衣原体浓度分别为10^(-9)、10^(-2)、10^(-3)、10^(-4),分组接种SPF鸡胚进行测定,检测了9株10^(-4)稀释的衣原体。并以B11001株衣原体试验性污染5批ND疫苗作检测。试验中取卵黄囊膜涂片,用中科院动物所提供的衣原体萤光抗体试剂盒进行检测,并辅以姬姆萨染色法检查,结果表明用荧光抗体染色可以检查到疫苗中衣原体或包涵体的存在,与姬姆萨染色法基本一致。
Abstract An immunofluorescence method for detecting the contamination of chlamydia in live vaccine was developed. Lots of Newcastle disease live vaccine that had been contaminated experimentally with Chlamydla psittaci B11001 in a rat o of l:10 ̄4 were neutralized with anti-Newcastle disease virus serum and then inoculated into SPF chicken embryos. Smears prepared from the yolk membranes of these infected embryos and stained with antichlamydia fluorescent antibodies produced specific flurescene under the vision of fluorescene microscope.These results were confirmed with the findings that observed in the giemsa's staining smears prepared from these same yolk membranes.
