MK-801对吗啡依赖大鼠戒断症状及中枢cAMP含量的影响 被引量:3

摘要 目的观察MK-801对吗啡依赖大鼠戒断症状及脊髓和脑干cAMP含量的影响。方法选择♂性SD大鼠32只,随机分为盐水对照组、吗啡依赖组、吗啡戒断组和MK-801组,每组8只。各组在纳洛酮激发戒断后立即观察戒断症状30min,纳洛酮激发戒断后1h,处死大鼠,分离脊髓和脑干,用放射免疫法检测cAMP的含量。结果MK-801组戒断症状总分为26.06±1.98,低于吗啡戒断组61.15±3.28(P<0.01)。MK-801组脊髓cAMP含量为0.8±0.26μmol.g-1,低于吗啡戒断组(P<0.05),而与吗啡依赖组相近(P>0.05);MK-801组脑干cAMP含量为1.62±0.38μmol.g-1,低于吗啡戒断组(P<0.05),而与吗啡依赖组相近(P>0.05)。结论MK-801减轻吗啡戒断症状的机制可能与其降低大鼠中枢cAMP的水平有关。 Aim To observe effects of MK-801 on withdrawal signs and cAMP of the spinal cord and brainstem in morphine dependence rats. Methods Thirty-two rats were divided into 4 groups (n = 8): control group, dependence group, withdrawal group and MK-801 group. Rats were killed 1 hour after naloxone challenge. Levels of cAMP in the spinal cord and brainstem were determined by radioimmunoassay. Result Total scores of withdrawal signs in the MK-801 group (26.06±1.98) were lower than those in the morphine withdrawal group (61.15±3.28) (P 〈 0.01). cAMP levels in the spinal cord (0.78±0.26 μmol·g^-1) and brainstem (1.62±0.38 μmol·g^-1) of MK-801 group were significantly lower than those in morphine withdrawal group (1.03±0.25 μmol·g^-1 and 2.06±0.41 μmol·g^-1) (p〈0.05) The difference was not statistically significant between the MK-801 group and the dependence group (P 〉 0.05). Conclusions Mechanisms underlying attenuation of morphine withdrawal by MK-801 may be that MK-801 decreases the level of cAMP in central nerves system.
