摘要
感觉神经元29KD蛋白的结构与功能研究,是神经科学领域中的一个重要方面。本实验从兔神经组织中分离、提取脊神经节感觉神经元特异蛋白,采用高效液相色谱二级分离法,纯化了感觉特异蛋白,电泳结果显示该蛋白的分子量为29KD,p17.8,经Waters氨基酸自动分析仪分析和Beckman LF-3200蛋白/多肽测序仪测试,揭示该蛋白的氨基酸组份及N端的15个氨基酸序列。研究结果显示,29KD蛋白是脊神经节感觉神经元特有的物质。
Revealed the functions and constructions of spinal sensory ganglion neurons from molecular level was an important role of neuron sciences. Sensory neuronal specific protein(SNSP) was isolated and purified from rabbit nervous tissues. And then the pure protein was prepared by the method of two-class separating of HPLC. SDS- polyacrylamide gel electrophoresis(PAGE) showed the SNSPband in the 29KD region, and isoelectric focusing electrophoresis indicated that pi is at pH7. 8. Anti-SNSP monoclonal antibody obtained by fusing SP2/0 myeloma cell wi^h splenic cells from mice immunized with the small cells specific protein, and then immunocytochemical studies show that the SNSP exists only in the fibers in small cells Ⅰ-Ⅱ lamina of spinal cord, dorsal roots, medium and small sized of ganglion and cutaneous branch of the peripheral nerves. Analysis of the N - terminal and compositions of amino acids, and immunocytochemical research suggested that the protein of 29KD may be the sensory neuronal specific protein of the spinal ganglion, which distribute, the way of superficial afferent fibers.
