摘要 为确定合适的采用流式细胞术计数水体中病毒的方法,本文比较了加入SYBR GreenⅠ染料后,不同染色孵育温度和染色孵育时间条件对病毒计数的影响。结果显示,样品染色孵育的温度应当控制在80℃左右,而染色时间不应少于10min。对样品进行固定后再染色测定,可以帮助染料迅速与病毒的核酸物质相结合,从而提高染色质量。 The comparisons were made between different time and temperature of incubation in water samples after staining with SYBR Green Ⅰ. A proper method was obtained for the enumerating marine viruses by flow cytometry. The results showed that the temperature for incubation should be at about 80℃, while the time for incubation should be no less than 10 min. The fixation of samples before stain may contribute to the quick combination of the dye and the nucleic acids of viruses, which would consequently improve the staining quality.
