摘要
目的应用双向电泳和质谱技术分析食管癌患者手术前后血清中蛋白质变化情况,寻找用于食管癌早期诊断的特异标志物。方法双向电泳分离正常人及食管癌患者手术前后血清总蛋白,银染显色;ImageMaster2-DE软件分析得到其蛋白表达谱。结果正常人及食管癌患者手术前后血清电泳平均筛选出的蛋白点数目分别为655、662、677;术前组与正常对照组蛋白点的密度和分布差异不明显,术后组与术前及正常组比较发现7个蛋白点存在明显差异:6个蛋白点表达上调,1个蛋白点表达下调。选取其中3个蛋白点进行MALDI-TOF-MS鉴定,结果显示此3个点的蛋白分别为血清淀粉样蛋白-A、血清淀粉样蛋白和触珠蛋白。结论血清淀粉样蛋白-A、触珠蛋白等可能与食管癌变和/或肿瘤手术本身有一定的相关性,它们与食管癌变的关系尚需更多的研究来证实;本研究为食管癌血清蛋白质组图谱的建立提供了有益的信息资料。
Objective To explore the specific biomarkers for early diagnosis by two-dimensional electrophoresis and mass spectrometry analysis from the same patient of EC before and after operation at high-incidence area for esophageal cancer ( EC ). Methods By two-dimensional electrophoresis, silver staining and imageMaster 2-DE analysis software, protein profiles were obtained in normal controls and patients before and after operation. Results Total 655,662 and 677 protein spots were identified respectively from eh normal controls and EC patients before and after operation. No significant difference of protein spots in density and distribution were identified with dramatic difference among the samples before and after operation. Sixprotein spots were up-regulated and one protein was down-regulated in groups after operation compared with those in normal control and the patients before operation. Three protein spots were selected to be characterized by MALDI-TOF-MS. The protein , from these three spots were identified as amyloid serum protein SAA, amyloid serum related protein and haptoglobin. Conclusion Amyloid serum protein SAA,amyloid serum related protein and haptoglobin might be related with EC and/or surgery by itself. The significance for these proteins needs tobe further characterized. The present study provides informative data for the establishment of serum proteinprofile related with EC.
