摘要
目的探讨低氧对机体外周血单个核细胞(PBM C)迁移的影响及其临床意义。方法分离外周血单个核细胞,采用T ransw e ll小室实验检测常氧和低氧条件下PBM C迁移能力的变化,流式细胞术(FCM)检测常氧和低氧条件下T、B淋巴细胞,NK细胞及单核细胞迁移的变化。结果低氧环境下PBM C的迁移率降低31%。对其细胞亚群的分析结果显示,NK细胞的迁移率降低31.7%,单核细胞的迁移率降低63.1%,T、B淋巴细胞的迁移率无显著变化。结论低氧能显著影响单核细胞及NK细胞的迁移功能;检测低氧条件下PBM C的迁移能力,可为以免疫细胞迁移功能为靶点的炎症等疾病提供新的治疗对策。
[Objective] To study the affect of hypoxia on migration capacity of human peripheral blood mononuclear cells (PBMC) and its biological significance. [Methods] PBMC were isolated and its migration capability were detected by Transwell system on normoxic and hypoxic condition. Then the migration of T and B lymphocyte ,NK cells and PBMC were analyzed by flow cytometry. [Results] The migration rate decreased 31% in hypoxia. The subtypes analysis showed that the migration rate of NK cell decreased 31.7% ,and mononuclear cell decreased 63.10%. But hypoxia had no affection both on T and B lymphocytes. [Conclusions] These results indicate that hypoxia can significantly affect the migration of monocytes and NK cells. Some diseases have therapy target such as immune cell migration ,and it may provide new therapy methods for those diseases by checking PBMC migration capability in hypoxia condition.
