水杉总黄酮对胰岛素样生长因子-Ⅰ诱导的乳鼠心肌细胞肥大的抑制作用 被引量:3

摘要 目的探讨水杉总黄酮(TFM)对胰岛素样生长因子-Ⅰ(IGF-Ⅰ)诱导的体外培养的乳鼠心肌细胞DNA和RNA合成的影响。方法体外分离培养乳鼠心肌细胞,分成4组,即正常对照组(C组)、IGF-Ⅰ组、IGF-Ⅰ+TFM组、IGF-Ⅰ+月见草油(OBL)阳性对照组,采用细胞CT扫描定量分析心肌细胞的DNA、RNA含量。结果IGF-Ⅰ组心肌细胞的DNA和RNA合成明显增加,与正常对照组比较,均有显著性差异(P<0.05);IGF-Ⅰ+TFM组心肌细胞的DNA和RNA合成明显低于IGF-Ⅰ组,两组比较差异明显(P<0.05),与C组接近(P>0.05)。结论水杉总黄酮对IGF-Ⅰ诱导的乳鼠心肌细胞的肥大具有抑制作用。 Objective:To investigate the effect of total Flavone of Metasequoia Mlyptostr-oboides on synthesizing of DNA and RNA of rat myocardial cells ( MCs). Methods: Isolated and cultured MCs of neonatal Sprague-Dawley (SD) were divided in to four groups, that is contral, insulin-like growth factor-Ⅰ ( IGF-Ⅰ), IGF-Ⅰ+ Total Flavone of Metasequoia Glyptostroboides(TFM) and IGF-Ⅰ+ Oenothera Biennis L oil(OBL) group. The DNA and RNA of MCs of all groups were measured by cell Computor tolo after incubation. Results: Comparing with the contral groups the content of DNA and RNA of MCs in IGFs group was siglificently increased ( P 〈 0.05), but the DNA and RNA of MCs in IGF-Ⅰ+ TFM and IGF-Ⅰ+ OBL groups obviously decreased( P 〈 0.05) and close to C group, Conclusion: TFM effectively inhibits the proliferation of cultured MCs in rat by IGF- I inducing, which may play a role in the regression of heart remodeling.
