摘要
采用95%(V/V)、50%(V/V)醇提荔枝核,经石油醚、乙酸乙酯、正丁醇依次萃取后,再利用硅胶柱层析等方法分离纯化,得到化合物Ⅰ,通过理化性质和波谱技术,鉴定为黄烷-3-醇类化合物表儿茶素。原花青素是由多羟基黄烷-3-醇单元构成的低聚体和多聚体,由不同数量的儿茶素或表儿茶素结合而成,通过单因素实验和L1(645)正交实验,确定原花青素的最佳提取工艺。最佳提取条件为:乙醇浓度50%,浸提温度60℃,料液比1∶20(W/V)、浸提时间3h。最佳条件下收率为5.05%。
The Litchi seed was extracted by 95 %,50 % ethanol. The extraction was successively partitioned with petroleum ether,EtOAc,n-BuOH; then the EtOAc fraction was separated by silica gel column chromatography, to afford the compound Ⅰ .On the basis of chemical and spectral analysis, the compound I was identified as (-)-epicatechin belongs to tlavan-3-ol. Base on this compound, the optimum extraction process of procyanidin was optimized with single factor test and L16 (45)orthogonal test. Optimum extraction conditions were determined as fol- lows: 50 %EtOH as extracting solvent, temperature 60 ℃,stock ratio 1:20(mass:volume),time 3 h.
