摘要
对硫酸软骨素的分光光度测定方法作了系统的研究,确定了其最佳水解条件为:样品在8mol/L盐酸:冰醋酸=2∶1的酸性溶液中水解60min,水解温度为100℃;水解液经间苯三酚试剂显色15min后进行光度分析。与已有分析方法比较,改进后的分析方法不仅操作条件稳定,而且测定精密度较高,重现性较好。
The conditions of the hydrolysis and photometric analysis of Chondroitin sulfate were studied. The op- timum photometric analysis conditions were confirmed. The samples were hydrolyzed under the condition of HCl (8 mol/L) and glacial with the ratio of 2:1 that was heated at 100 ℃ for 60 min. The phloroglucin reagent was added to this hydrolysis solution which was determined after 15 min. This method is simple, fast, accurate and convenient. it is one of the reliable means for Chondroitin sulfate determination.
