缬沙坦对兔动脉成形术后血管内膜增生及ICAM-1表达的影响

摘要 目的观察缬沙坦对兔动脉成形术后血管内膜增生及细胞间黏附分子-1(ICAM-1)表达的影响。方法雄性新西兰白兔24只,随机分成三组,对照组给予普通饲料12周;模型组和缬沙坦组予高胆固醇喂养8周后行球囊成形术;缬沙坦组给予缬沙坦干预治疗4周。三组均取腹主动脉行病理形态学观察及ICAM-1免疫组织化学分析。结果缬沙坦组较模型组内膜厚度及面积明显减小(P<0.01)。免疫组织化学分析显示,缬沙坦组与模型组相比ICAM-1表达显著减少(P<0.01)。结论缬沙坦可以显著减轻兔腹主动脉成形术后内膜增生,其机制可能与抑制血管内膜ICAM-1表达,从而抑制损伤内膜炎症反应有关。 [Objective] To investigate the effect of valsartan on neointimal proliferation and expression of intercellular adhesion molecule-1 (ICAM-1) after balloon angioplasty in rabbits. [Methods]24 male New Zealand white rabbits were randomly divided into three groups. The control group was fed with common diet. The model group and the valsartan group were fed with hypercholesterolemia diet for 4 weeks, then they were underwent ballon angioplasty. During 4 weeks after operation, the model group was fed with common diet, whereas the valsartan group was fed with the admixture of valsartan and common diet. The abdominal aorta was examined with pathologic and morphologic analysis, and expression of ICAM-1 was analyzed with immunohistochemical method. [Results] Compared with the model group, the neointimal thickness and area decreased by 56.58% and 66.81% respectively in the valsartan group. ICAM-1 expression in valsartan group was significantly lower than that in model group. [Conclusions] Valsartan can inhibit neointimal proliferation of rabbit's abdominal aorta after balloon angioplasty, the mechanism might be correlated with its reducing expression of ICAM-1, and decreasing inflammation of vascular injury.
