摘要
为了建立简便实用的小麦Wx蛋白基因的分子标记辅助选择体系,采用6对SSR和STS引物对16个已知Wx蛋白类型的小麦品种(系)进行分子标记检测的验证和比较,筛选出可采用同一PCR扩增程序、在琼脂糖凝胶上分离扩增产物、分别检测3个Wx基因的3对引物。利用这些引物对扬麦158与EH-5的杂交F2代进行Wx基因型检测,获得全部27种基因型,表明这些引物可以有效用于Wx蛋白基因的分子标记辅助选择。
In order to establish a simple and practical marker-assisted system for breeding of Wx genotype of wheat, six pairs of SSR or STS primers were employed to detect Wx genes of 16 wheat varieties whose Wx protein types genes had been identified by SDS-PAGE. The result showed that each of 3 primer pairs could identify one of the 3 Wx genes by the same PCR procedure, respectively, and the fragments of PCR could be separated in agarose gel. The F2 individuals derived from the cross of Yangmai 158 and EH-5 were analyzed by these primers and 27 genotypes of Wx genes were indentified. This indicated that these primers could be efficiently used for marker-assisted selection of Wx genes in the future.
