摘要
目的探讨功能性和非功能性肾上腺肿瘤组织中Bcl-2的表达水平和细胞凋亡的关系。方法运用免疫组织化学染色和TUNEL法检测细胞凋亡情况,探讨4例正常肾上腺(NA)、33例有功能性肾上腺肿瘤(FAT)和23例非功能性肾上腺肿瘤(NFAT)的Bcl-2表达及细胞凋亡情况。结果Bcl-2阳性细胞的平均百分数在NA、FAT、NFAT分别为(3.8±1.1)%、(6.3±1.2)%、(13.1±1.8)%,其中FAT与NFAT、NA与NFAT比较,均有显著性差异(P<0.05);FAT与NA比较无显著性差异(P>0.05)。凋亡的阳性细胞率FAT(1.14±0.30)%高于NFAT的(0.48±0.25)%和NA(0.18±0.05)%,其中FAT与NFAT、FAT与NA、NFAT与NA比较均有显著性差异(P<0.05)。Bcl-2的表达与细胞凋亡指数(AI)呈显著负相关(rs=-0.560,P<0.02;rs=-0.530,P<0.03)。结论Bcl-2表达与细胞凋亡抑制关系密切;Bcl-2的表达及细胞凋亡检测对功能性肾上腺肿瘤和非功能性肾上腺肿瘤有一定诊断意义。
Objective To explore the correlation of expression of bcl-2 protein and cell apoptosis in functioning and non- functioning adrenal tumours. Methods Formalin-fixed and paraffin-embedded material, comprising 33 cases of functioning adrenal tumours (FAT), 23 non-functioning adrenal tumours (NFAT) and 4 normal adrenal gland (NA) tissue, were analyzed by immunohistochemical staining to detect the expression of Bcl-2 and cell apoptosis in TUNEL. Results FAT (6.3 ± 1.2) % and normal adrenal gland (3.8 ± 1.1 )% groups showed an lower average percentage of Bcl-2 positive cells than NFAT (13.1 ± 1.8)% group( P〈0. 05 by SAS ). The rate of apoptotic cells was higher in FAT (1.14±0.30)% than in NFAT (0.48± 0.25) % and in normal adrenal gland (0. 18±0.05)% (P〈0.05 by SAS). There were negative correlations between positive rate of bcl-2 protein expression and the apoptotic index. Conclusion Immunohistochemical staining detecting the expression of Bcl-2 and cell apoptosis in TUNEL appears to be useful in differentiating FAT from NFAT. Overexpression of bcl-2 protein is associated with depression of apoptosis.
