摘要
目的:研究药物API0134对实验性急性心肌缺血—再灌注过程中血浆6-酮-前列腺素F1α和粒细胞产生氧自由基的影响。方法:结扎犬冠状动脉左前降支90分钟后行再灌注120分钟。用药组(n=8)在冠状动脉左前降支结扎45分钟时静脉给予API0134(9mg/kg)至再灌注60分钟。对照组(n=8)给予同等量5%葡萄糖盐水。放射免疫法测定血浆6-酮-前列腺素F1α和凝血烷B2(TXB2,血栓素B2)含量,化学发光法测定粒细胞产生氧自由基的量。结果:与对照组比较,用药组在用药后心肌缺血—再灌注期间,血6-酮-前列腺素F1α增高(P<0.05)、凝血烷B2降低,粒细胞产生氧自由基的量减少(P<0.05)。结论:API0134可促进心肌缺血—再灌注过程中6-酮-前列腺素F1α(即前列腺素)的生成,抑制粒细胞产生氧自由基,这可能是该药减轻心肌再灌注损伤的重要机制之一。
Objective:To study the effects of Andrographi Peniculata isolates 0134 ( API 0134 )on plasma 6 keto prostaglandin(PG)F 1α and the release of oxygen free radicals by neutrophils during experimental acute myocardial ischemia reperfusion. Methods:Ligation of the left anterior descending coronary artery (LAD) in dog lasted 90 min followed by 120 min reperfusion.In the treated group ( n =8),infusion of API 0134 (9 mg/kg) started at 45 min after LAD ligation,and lasted to 60 min after reperfusion.In the control group ( n =8),the same amount of 5% glucose saline was given at the same time as above.Plasma concentrations of 6 keto PGF 1α and thromboxane(TXB 2) were determined by radioimmunoassay.Oxygen free radicals generated by neutrophils was measured by chemiluminescence. Results:Compared with the control group,plasma 6 keto PGF 1α in the treated group was increased ( p <0 05),TXB 2 decreased,and neutrophil induced oxygen free radicals were inhibited( p <0 05),during ischemia reperfusion after administration of API 0134 . Conclusion: API 0134 may increase the production of plasma 6 keto PGF 1α ,inhibit neutrophil induced oxygen free radicals during myocardial ischemia and reperfusion,which may be one of the important mechanisms in alleviating myocardial reperfusion injury.
