摘要
成骨细胞样细胞体外培养^(14)C-脯氨酸掺入液闪测定及放射自显影(ARG)观察显示:鸡胚额骨分离细胞原代培养3d就出现比较高的^(14)C-脯氨酸的掺入值,峰值出现在培养7d,细胞外^(14)C-脯氨酸掺入最大值出现在培养10d并持续一定时间,这与细胞内胶原合成及分泌有密切的关系。结果显示体外培养成骨细胞样细胞较早就有胶原合成;细胞增殖和细胞胶原合成同时发生,且细胞内胶原合成率在细胞的对数增殖期最高。该实验为今后进一步研究药物对成骨细胞样细胞体外培养实验中^(14)C-脯氨酸掺入提供最佳参考时间。
Experimental study on14C-proline incorporation into osteoblast-like cells from embryonic leghorn chicken frontal bone cultured in vitro was carried out with liquid scintillation counting method and radioautographic technique。It was found that relatively high 14C-proline incorporation into osteoblast-like cells took peak at 3rd day of culture but the peak was on the 7th day of culture。In the extra-cellular matrix,the maximum value of 14C-proline incorporation appeared on the 10~13 days of culture,as a result of formation and secretion of the intercellular collagen。The results revealed that:(1)the isolated osteoblast-like cells can produce collagen in the early stage of culture.(2)the proliferation of cells and the collagen synthesis by the cells proceed at the same time.(3)the peak value of the collagen formation was at the interval logarithinic proliferation of the cells。This experiment provided appropriate time of 14C-proline incorporation for future studying drug activity on cultured osteoblast-like cells。
