摘要 目的通过检测肾脏TGFβ-1mRNA的表达及其病理变化,研究肝细胞生长因子(HGF)对2型糖尿病肾病不同阶段的治疗作用。方法雄性Wistar大鼠随机分为对照组、糖尿病肾病组、治疗组(HGF组),HGF组分别于注射链脲佐菌素后2个月和6个月时给予HGF 500μg/(kg.d),连续1个月后处死。取一侧肾脏HE染色作病理切片,另一侧肾脏用逆转录多聚酶链反应(RT-PCR)检测TGFβ-1mRNA的表达。结果HGF能明显抑制糖尿病大鼠肾脏TGFβ-1mRNA表达的上调。结论HGF可抑制早期糖尿病肾病进一步发展,减轻临床糖尿病肾病的蛋白尿,使肾小球基膜(GBM)明显变薄和细胞外基质降解从而改善并逆转临床糖尿病肾病大鼠肾脏的功能和形态的变化。 Objective To research the effect of HGF on diabetes nephropathy by detecting the expression of TGF-β1 mRNA and the morphologic changes in nephropathy. Methods Male Wistar rats were divided randomly into three groups: normal control group ( NC ), diabetic nephropathy(DN ) and therapy group (HGF). For HGF group, hHGF[ 500 μg/(kg·d) ] was injected for one month in months 2 and 6 after injecting STZ,and then the rats were killed. The right kidney samples were used to morphologic analysis and the left kidney samples were used to measure the expression of TGF-β1mRNA by RT-PCR. Result HGF could inhibit the up-regulation of the expression of TGF-β1mRNA in diabetic nephropathy. Conclusion HGF can inhibit the progress of DN in the early stage. HGF can decrease urine albumine, make glomerular basement membrane (GBM) thinner and extracellular material(ECM)degrading. So it can improve and reverse the changes in function and form of the Wister rats' kidney suffering from clinical diabetes.
