SLC基因修饰瘤苗免疫小鼠脾细胞过继治疗胃癌实验研究

摘要 目的探讨次级淋巴组织趋化因子(secondarylymphoidtissuechemokine,SLC)免疫基因转染胃癌细胞瘤苗免疫小鼠后获得的脾淋巴细胞后治疗经同一免疫基因修饰瘤苗转染的小鼠胃癌的可行性和疗效。方法用200PFU/细胞滴度的重组腺病毒载体(pVAX1)将人SLC基因转染小鼠胃癌SGC-7901细胞株,经80mg/L丝裂霉素(MMC)处理制备成胃癌细胞瘤苗,免疫同系小鼠后分离其脾淋巴细胞,转染SLC基因(腺病毒载体滴度200PFU/细胞)后注射入直径1cm的小鼠胃癌淋巴结转移模型瘤体内,观察抗瘤效果。结果转SLC基因瘤苗治疗组小鼠瘤体增加值最小(0.08±0.05)cm3,与各对照组比差异有显著性[转染SLC基因,未转染SLC和生理盐水注射组分别为(1.46±0.15)、(1.56±0.23)和(2.12±0.54)cm3,P<0.05]。结论SLC基因修饰癌瘤苗诱导小鼠产生的免疫脾淋巴细胞可能成为一种新的过继免疫治疗效应细胞及SLC基因瘤苗诱导特异性主动免疫治疗结合将有更优越的抗瘤效果和应用前景。 Objective To find the feasibility and antitumor effect of mSLC gene transfected splenocytes derived from SLC geneco - transfected gastric cancer vaccinersimmunized nude mice. Methods The murine gastric cancer cell strain SGH - 7901 transfected mouse SLC gene via recombinant adenovines vectors (200PFU/cell) were treated with 80mg/L mytomycin (MMC) to prepare gastric cancer cell vaccines: The spleenocytes isolated from the mice immunized with these vaccines were transfected with SLC gene or nontransfected and injected into the mice xenograft gastric cancer with tumor diameter being 1 cm) respectively, The adoptive antitumor effect was investigated, Results The increasing volume of the tumor injected with SLC gene transfected splenocytes intratumorally was significantly smaller than in the control groupe (The increasing volume of the tumor in the SLC gene trsfectec, nontransfected splenocytes and saline injected groupes was (1,46 ±0.15) ,(1.56 ±0.23 )and (2.12 ±0.54)cm^3 respectively ,P 〈0. 05. Conclusions Significant tumor inhibitory effect was demonstrated in the immunized - activated spleenocytes derived from SLC gene transfected gastric cancer vaccine immunized mice and was further improved while modified with SLC gene . It is suggested that it would be more efficiency if gene therapy ,specific active immunotherapy in combination with adoptive cellular immunotherapy were adopted.
