摘要
目的探讨糖尿病大鼠肾皮质中转化生长因子β1(TGF-β1)、波形蛋白(vimentin)和结蛋白(desmin)表达的变化规律及其相互关系。方法建立STZ诱导的SD大鼠糖尿病模型;用RT-PCR法检测造模成功后第3、7、14、30、60和90天糖尿病大鼠肾皮质中TGF-β1、vimentin和desminmRNA的表达水平。结果①糖尿病大鼠肾皮质TGF-β1和vimentinmRNA的表达分别从模型建立后第7天和第14天起逐渐增多,desminmRNA的表达从第14天起却逐渐减少,这种变化持续至整个实验期末,糖尿病大鼠和对照组大鼠相比差异具有显著性;②糖尿病大鼠肾皮质TGF-β1和vimentin的mRNA表达呈显著正相关,TGF-β1和desmin的mRNA表达却呈显著负相关,desmin和vimentin的mRNA表达也呈显著负相关。结论糖尿病大鼠TGF-β1在肾皮质局部表达增多可能促进肾小管上皮细胞向成纤维细胞转化。
Objective To study the expression of TGF - β1, vimentin and desmin gene in the renal cortex of STZ induced diabetic rats. Methods Diabetes was induced in 32 male SD rats by single intraperitoneal injection of 1.0% STZ (70mg/kg). 36 age and weight, sex matched SD rats used as controls. The expression of TGF - β1, vimentin and desmin mRNA in the renal cortex were detected by RT - PCR on the 3^rd , the 7^th , the 14^th , the 30^th , the 60^th and the 90^th days after the DM rat model established. Results ① The expression of TGF - β1 , vimentin mRNA in the renal cortex of diabetic rats gradually increased respectively from the 7^th day and the 14^th day after the model established, and the expressive intensity was significantly greater than that in controls (P 〈 0.05 or P 〈 0.01 ). However, the expression of desmin mRNA in the renal cortex of diabetic rats gradually decreased from the 14^th day after the model established, and the expressive intensity was significantly less than that in controls ( P 〈 0.05 or P 〈 0.01 ). ② The expression level of TGF - β1 mRNA correlated positively to that of vimentin mRNA in the renal cortex of diabetic rats, while the expression level of desmin mRNA correlated negatively to that of TGF - β1 mRNA and to that of vimentin mRNA in the renal cortex of diabetic rats. Conclusions There may be partial transformation of renal tubular epithelial into fibroblast and TGF - β may play a role in this transformation during the progressive interstitial fibrosis of diabetic nephropathy.
