皮肤组织工程种子细胞-表皮干细胞提供来源选择的研究

Selecting a Better Donor Site for the Epidermal Cell Source of Skin Tissue Engineering

目的寻找表皮基底层细胞中含有较高干细胞比例的供皮区。方法选择不同供皮部位，利用免疫组织化学的方法，以K19和整合素β1作为表皮干细胞鉴定标记。结果皮肤基底层表皮干细胞以包皮及阴囊最多，其次是臀部，背部多于胸部。四肢近端外侧多于四肢近端内侧，头皮、手掌及足底皮肤基底层干细胞数量较少。结论从组织工程种子细胞的供应来源考虑，以男性包皮最佳，在客观上也容易获得。

Objective To select a human skin part which has a higher ratio of stem cell inside the basal layer as the donor site for skin tissue engineering. Methods Different skin parts were obtained from adult volunteers. Epidermal stem cell was identified by using K19 and integrin β1 as the biochemical markers. Results The ratios of K19 and integrin β1 positive cells were different in the different skin part. The foreskin and scrotum sections have the most, next was hip. Posterior trunk was higher than anterior trunk. Outer side of thigh and upper arm was higher than anterior trunk. Outer side of thigh and upper arm was higher than inner side of thigh and upper arm. The ratio was lowest at scalp, palm and vola. Conclusion The foreskin was the best choice as the donor for skin tissue engineering.
