摘要
目的探讨血清在绿脓杆菌和角膜基质细胞介导的角膜胶原降解时的作用。方法Ⅰ型胶原凝胶,不混悬及混悬角膜基质细胞,在不同的血清浓度下(0、5%、10%、20%)与绿脓杆菌培养液共同培养24h,超滤去除天然原纤维,超滤液经过水解,分光光度计测定其羟脯氨酸(HYP)的量作为胶原降解的活性单位。结果不混悬角膜基质细胞组,随血清浓度增加绿脓杆菌所引起的胶原降解量无变化,差异无显著性(P>0.05);混悬角膜基质细胞组,随着血清浓度增加绿脓杆菌所引起的胶原降解量减少,差异有显著性(P<0.025orP<0.05);在相同的血清浓度(0、5%、10%)时,混悬角膜基质细胞组胶原降解量高于不混悬角膜基质细胞组,差异有显著性(P<0.025orP<0.05)。结论血清中的某些活性成分可抑制绿脓杆菌诱导的角膜基质细胞胶原降解。
Objective To investigate the effects of serum on degradation of keratecyte-collagen induced by Pseudomonas pyocyanea and keratecyte.Methods The keratocyte-collagen gels were incubated with Pseudomonas pyecyanea and without (group Ⅰ )or with(group Ⅱ ) keratecyte and different concentration of serum (0,5% ,10% ,20% ) in MEM culture medium separately 24 hours later, to evaluate the collagen degradation level, the amount of hydroxyproline(HYP) were measured. Results In the group Ⅰ ,the amount of HYP by Pseudomonas pyecyanea was unchanged with the increasing of serum concentration,There were not significant differences(P 〉 0.05) ; In the group Ⅱ ,the amount of HYP by Pseudomonas pyocyanea was decreased with the increasing of serum concentration,There were significant differences( P 〈 0.025 or P 〈 0.05) ;in the identical of serum concentration (0.5%, 10% ), the amount of HYP by Pseudomonas pyecyanea was decreased, There were significant differences( P 〈 0. 025 or P 〈 0.05). Conclusion The some composition of serum inhibites keratocyte-collagen degradation induced by Pseudemonas pyecyanea.
