摘要
目的建立PBMC穿孔素活性的测定方法并探讨其对肺癌转移预报的价值。方法以PBMC为效应细胞、兔红细胞为靶细胞,根据Hb的过氧化物酶样作用,以OPD为显色底物测定靶细胞裂解后释出的Hb的多少,从而反映穿孔素溶血活性大小。并以此指标检测肺癌确诊病人。结果效靶比1∶20,反应体系中加入Ca2+,5%CO2,37℃孵育16小时,可较好反映穿孔素的溶血活性。肺癌患者的PBMC穿孔素溶血活性较正常人明显降低,有肿瘤转移者较无转移者降低。结论穿孔素溶血活性的测定对肺癌的诊断及癌肿转移的预报具有重要价值。
Objective To establish the detection of the hemolytic activity of Perforin and to study the predictive action of Perforin at metastasis of pulmonary carcinoma. Methods Rabbit's red cells were used as target cells and PBMC as effect cells in this test. The quantity of released Hb from the broken target cells was examined according to the action of Hb as peroxidase, thus the activity of Perforin was reflected. The patients of pulmonary carcinoma were examined with this experimentation. Results The hemolytic activity of Perforin could be detected well under these conditions: effect: target = 1 : 20; there was Ca^2 + in reaction system and incubation for 16 hours in 37℃ ,5% CO2 incubator. Compared with normal person, the hemolytic activity of Perfofin in pulmonary carcinoma was significantly redactive and more redactive in the patients with metastasis of pulmonary carcinoma. Conelusions The detection of hemolytic activity of Perforin was useful to the diagnosis of pulmonary carcinoma and the prediction of metastasis of pulmonary carcinoma.
