摘要
以麸皮为主要原料,添加少量稻壳以增加疏松度,加入与物料同重量的水,混匀,经灭菌、冷却后,接种诱变后的黑曲霉3.324菌株。采用曲盘固体发酵法,在保持室温28-30℃,干温相差1℃的条件下发酵6 d,经生化方法提纯得到植酸酶精制品,每克干曲(以麸皮原料计算)产植酸酶约0.145 g,精制品植酸酶活力达300 U.g-1。
Using of bran as main raw material, and a low amount of rice hull was added to increase the sedimentation of raw materials. The same amount of water was mixed equally with the raw materials. After sterilizing and cooling, the aspergillus niger was inoculated to the processed materials. A solid fermentation at the temperature of 28 - 30 ℃, and temperature difference of 1 ℃ was employed and lasted for 6 days. The amount of produced phytase was 0.145 g per kg, and the enzyme activity of purified phyatse was detected at a level of 300 U·g^-1.
