姜黄素对血管平滑肌细胞源性荷脂细胞胆固醇代谢及SREBP-1表达的影响 被引量:6

摘要 目的观察姜黄素对血管平滑肌细胞源性荷脂细胞胆固醇代谢的影响,并初步探讨SREBP-1蛋白表达在其中的作用。方法以血管平滑肌细胞源性荷脂细胞为模型,不同浓度姜黄素(12.5、25.0、50.0μmol/L)作用于细胞24 h及25.0μmol/L姜黄素不同时间(0、6、12、24、48 h)作用于细胞,高效液相色谱分析法检测细胞内总胆固醇。游离胆固醇和胆固醇酯的含量。在SREBP-1蛋白抑制剂ALLN干预下,25.0μmol/L姜黄素处理平滑肌细胞源性荷脂细胞24 h,Western印迹法检测SREBP-1蛋白的表达。结果不同浓度姜黄素(12.5、25.0、50.0μmol/L)作用于血管平滑肌细胞源性荷脂细胞24 h及25.0μmol/L姜黄素不同时间(06、1、22、4、48 h)作用于血管平滑肌细胞源性荷脂细胞,细胞内总胆固醇,游离胆固醇和胆固醇酯含量呈现一定的剂量和时间依赖性下降趋势;在SREBP-1蛋白抑制剂ALLN干预下,25.0μmol/L姜黄素处理平滑肌细胞源性荷脂细胞24 h,SREBP-1表达下调,总胆固醇、游离胆固醇和胆固醇酯含量无明显变化。结论姜黄素能引起血管平滑肌细胞源性荷脂细胞总胆固醇、游离胆固醇和胆固醇酯含量下降,且这种作用跟SREBP-1蛋白的表达上调有一定关系。 Objective To observe the effect of curcumin on the cholesterol metabolism in vascular smooth muscle cell - derived cholesterol - road cell, and investigate the effct of SREBP - 1 at the process. Methods using vascular smooth muscle cell - derived cholesterol - road cell as investigating object, curcumin of different density (0,12.5,25.0,50.0 μmol/L for 24 hours)and different time(0,6, 12,24,48 h of 25.0 μmol/L) effects on cells, and using high performance liquid phase analytic process detects the level of total, free and esterified cholesterol. curcumin of 25.0 μmol/L effects on cells with the intervention of SREBP - 1 suppressive agent ALLAN, and using Western Blot detects the expression of SREBP-1. Results curcumin of different density(0, 12.5,25.0,50.0 μmol/L for 24 hours) and different time (0,6, 12,24,48 h of 25.0 μmol/L) effects on vascular smooth muscle cell -derived cholesterol - road cells, the level of total, free and esterified cholesterol decrease in cells , and the change is shown for dose and time dependence, curcumin of 25.0μmol/L effects on cells with the intervention of SREBP - 1 suppressive agent ALLN, SREBP- 1 expression is down - regulated, and no marked change is observed at the level of total,free and esterified cholesterol in cells. Conclusion curcumin can degrade the level of total,free and esterified cholesterol in vascular smooth muscle cell- derived cholesterol- road cells, and it is contributed to the up- regulation of SREBP - 1 expression at some degree.
