免疫转印技术分析流行性出血热病毒抗原组分的研究

ANALYSIS ON EHFV COMPONENT ANTIGEN BY IMMUNOBLOT TECHNIQUE

免疫转印技术分析流行性出血热病毒（ＥＨＦＶ）陈株、Ｒ_(２２)株结构蛋白。病毒抗原系感染乳鼠脑内增殖，硫酸鱼精蛋白—ＰＥＧ法提取。采用免疫ＥＨＦＶ血清（浙江Ｒ３血清，抗Ｒ２２血清）及抗ＥＨＦＶ单克隆抗体（Ｒ_(３１)、Ａ_(１９)、Ａ_(２０)、Ａ_(３５)、Ａ_(２５—１)）对两株病毒多肽抗原进行分析。结果陈株具有７５ＫＤ，６９ＫＤ，６６ＫＤ，６１ＫＤ４条多肽，Ｒ２２株具有７２ＫＤ．６９ＫＤ，的ＫＤ３条多肽。抗体识别模式表明两株病毒抗原仅有微小的株间差异。

mmunobloting analysis was camed out on the virion protein of strain chen and strain R22viruses,the a eti ologic agent of EHF.The viral antigen was purified and characterized roughly byprotamine sulfate and polyethylene glycol precipition from brain tissue of suckling mice infectedwith EHFV strain chen or strain R22.Immunobloting analysis was performed withanti-EHFV sera(anti-R22 virus sera,Zhejian R3sera)and Mcab A19,A20,A35,A25-1,R31.Itshowed four bands on NC with molecular weights of 75KD,69KD,61KD,in strain chen and three bands on NC with molecular weights of 72KD,69KD,64KD in strain R22.All antibodiesreacted with the viral antigen of the two viruses.It shows a little difference between stra in clien and R22 in Virral antigen.
