PSD-007单体光敏剂对三株人癌细胞的光动力效应 被引量:1

PHOTODYNAMIC EFFECT OF THE PHOTOSENSITIZER PSD -007 ON THREE HUMAN TUMOR CELL LINES

摘要 PSD-007单体是已确定结构式的血卟啉类激光光敏剂。我们用人大肠癌细胞株SW1116,宫颈癌细胞株HeLa和红白血病细胞株K562,以北京产的同类光敏剂HPD作为药物对照,在波长630nm的铜蒸汽激光照射4J/cm2能量密度后,用MTT法测定两种光敏剂对3个人癌细胞的9个浓度组,计算50%细胞抑制率(IC50)和细胞生存率的均数,比较两个光敏剂对三株人癌细胞的光动力杀伤效应。实验结果显示:用K562细胞HPD的IC50为4mg/L,PSD-007为4mg/L;HeLa细胞IC50为4.5mg/L和4mg/L;SW1116细胞的IC50为4.5mg/L和1mg/L。在对两种药物7个不同浓度研究组中,抑瘤结果表明,各组间有显著性差异(P<0.05),说明随着药物浓度的增高光动力杀伤效应增大。 The structure formula of single PSD 007 a photosensitizer of hematoporphyrinoid,was definited and its photodynamic effects on three human tumor cell lines (SW1116,HeLa and K562) were measured with MTT assay.Three cell lines were treated with 9 concentration of PSD007 and HPD, subsequently subjected to an irradiation of a pulsed copper vapour laser at wavelength of 630nm and a fluence of 4 J/ cm2.As a control,three cell lines were treated with HPD (made in Peking).The IC50 and the means of cell survival rate were calculated to compare the photodynamic effect between cell lines of each group. The results were as following.(1) The cell lines of K562,HeLa and SW1116 teated with HPD,IC,.value were 4 mg/L,4.5 mg/L and 4. 5 mg/L respectively,treated with PSD-007,IC50 value were 4 mg/L,4 mg/L and 1mg/L separately.(2)At a seven points sery of drug concentrations,ranging from 8 mg/ L to 4 mg/ L,to each cell line,the tested tumor inhibitive rates to each drug point were different, and the P value between two of each group was less than 0.05, which suggested the efficacy of the photodynamic effect got bigger along with the concentration elevated in vitro.
