饲用酶制剂中羧甲基纤维素酶等5种酶活力的测定方法研究 被引量:13

摘要 利用3,5-二硝基水杨酸(DNS)比色法,对饲用酶制剂中羧甲基纤维素酶(Cx酶)、木聚糖酶、β-葡聚糖酶、β-葡萄糖苷酶和果胶酶5种酶活力的测定进行了研究.对DNS比色法测定5种酶活力的影响因素:底物水解的pH值、温度及时间等进行了选择试验,优化了分析条件.5种酶活力重复测定的相对标准偏差分别为6.22%~6.50%、7.90%~10.6%、8.70%~13.9%、3.00%~3.76%和7.11%. The activity of 5 enzymes such as carboxymethyl cellulose ( Cx enzyme) , xylanase, β-dextranase, β-glycosidase and petinase in feed enzyme preparations was determined by 3,5 - dinitrosalicylic acid (DNS) colorimetry. The selective experiments for factors influencing the activity of 5 enzymes such as Cx enzyme and etc. determined by DNS colorimetry, i.e. pH value of substrate hydrolysis, hydrolysis temperature and hydrolysis time were carried out, and the analysis conditions were optimized. The results showed that the relative standard deviation (RSD) of repeatedly determined activity of 5 enzymes were 6.22% - 6.50% , 7.90% - 10.6%, 8.70% N 13.9% , 3.00% N3.76% and 7.11%.
