摘要
目的通过检测子痫前期患者胎盘组织中TGFβ1和MMP-9(明胶酶B)的表达,探讨其在子痫前期发病机制中的作用及两者之间的相关性。方法用SP免疫组化染色法观察正常妊娠(15例),子痫前期(24例)剖宫产后胎盘组织中TGFβ1和MMP-9的分布与定位,通过HMIAS-2000型全自动医学彩色图像分析系统对其进行定量分析。结果在重度子痫前期组胎盘组织中TGFβ1表达明显高于正常对照组和轻度组(P<0.05),轻度组与正常组比较差异无显著性(P=0.087);MMP-9在轻、重子痫前期组胎盘组织中表达均明显低于正常对照组(P<0.05);胎盘组织中TGFβ1的表达水平和MMP-9的表达水平呈负相关(P<0.05)。结论胎盘组织中TGFβ1、MMP-9表达异常引起滋养细胞浸润不足,提示两者可能并共同参与子痫前期的发生和发展。
Objective: Study on expression of TGFβ1 and MMP -9 in the placenta tissues in order to evaluate the role of them and their relationship in preeclampsia. Methods : Immunohistochemistry was used on the normal group ( 15 cases) and the preeclampsia group (24 cases in total) to observe the location and distribution. High resolution pathological image analysis system was used to determine the expression of TGFβ1 and MMP -9. Results: The levels of TGFβ1 expression in the severe preeclampsia group were signif- icantly higher than those of normal and the slight group ( P 〈0. 05), and the expression of TGFβ1 was no significant difference between the slight preeclampsia and normal women ( P = 0. 087) The levels of MMP- 9 expression in the severe and slight preeclamp- sia group were significantly lower than those of normal group ( P 〈0. 05). There was a significantly negative correlation between the expression of TGFβ1 and MMP -9 in placental tissue ( P 〈0. 05). Conclusion: The abnormal expression of TGFβ1 and MMP -9 in placenta leads to insufficient trophoblasts infiltration and might be involved in the pathogenesis of preeclampsia.
