摘要
目的在体外实验中,探讨三氧化二砷(As2O3)诱导卵巢癌细胞发生凋亡的可能性,揭示该凋亡发生与bcl-2和bax之间的关系。方法采用TUNEL染色法,定性、定量地研究As2O3与卵巢癌细胞凋亡的关系;通过免疫组织化学法检测凋亡相关基因bcl-2和bax的表达。结果As2O3在体外能诱导卵巢癌细胞发生凋亡,下调bcl-2的表达,增强bax的表达。结论诱导卵巢癌细胞发生凋亡是As2O3抗卵巢癌作用的机制之一,As2O3可能通过下调bcl-2的表达及增强bax的表达诱导卵巢癌细胞发生凋亡。
Objective: To investigate the apoptosis in ovarian cancer cells induced by As2O3 , and the relation between this apoptosis and expression of bel - 2 and bax. Methods : In vitro experiments, TUNEL staining method were used to quantitatively and qualitively detect the apoptosis status of ovarian cancer cell line A2780 before and after the As2O3 treatment. Immunohistoehemieal staining were used to detect the expression of apoptosis -regulated gene bel -2 and bax. Results : As2O3 was able to induce the apoptosis in ovarian eaneer. As2O3 can reduce the expression of apoptosis - regulated gene bel - 2, and increase the expression of apoptosis - regulated gene bax. Conclusion: As2O3 is able to induce the apoptosis in ovarian cancer. This apoptosis may be mediated by down - regulation of apoptosis - regulated gene bel - 2 and up - regulation of bax.
