细胞间黏附分子-1在桥本甲状腺炎中的表达及其影响因素 被引量:2

摘要 目的检测桥本甲状腺炎患者甲状腺组织中细胞间黏附分子-1(CD54)的表达和离体的甲状腺滤泡上皮细胞在不同刺激因子作用下CD54表达水平的变化。方法对41例桥本甲状腺炎组织采用免疫组化S-P法染色,图像分析系统对染色强度进行定量分析测定CD54在甲状腺组织中阳性表达率和表达面积。经过分离培养的甲状腺细胞,在不同浓度IL-1β和NaI干预下,采用流式细胞仪检测其CD54表达的变化。结果桥本甲状腺炎CD54阳性表达率明显高于正常对照组(P<0.05);其阳性面积也明显高于正常对照组(P<0.05)。离体的桥本甲状腺炎甲状腺细胞CD54表达水平虽然较正常对照高,但并无显著差异;桥本甲状腺炎细胞在不同干预因素下CD54表达水平都明显高于正常对照组(P<0.05);在IL-1β干预下,发现高水平的IL-1β可以使正常甲状腺细胞CD54的表达明显升高,但桥本甲状腺炎甲状腺细胞对IL-1β的敏感性更强,低剂量的IL-1β可以显著增加CD54的表达,加入NaI后CD54表达明显增加。结论CD54在桥本甲状腺炎中的升高可能是一种继发反应,与IL-1β和NaI有关,在桥本甲状腺炎的发病机制中起重要作用。 Objective To dectect the expression of CD54 in thyroid tissue of Hashimotos thyroiditis patients, and expression changes of CD54 on thyroid cells interferred by different agents. Methods The thyroid tissues from 41 cases of Hashimotos thyroiditis were collected and 26 normal thyroid tissues served as normal controls. All thyroid tissues were identified by pathological examination. The positive expression rate and area of CD54 were investigated by immunohistochemical method and quantitative analysis of image analysis system in all thyroid tissues. The expression changes of CD54 in the isolated thyroid cells interferred by 100,500, 1 000 pg/ ml IL-1β or 10 mg/L NaI or 1 000 pg/ml IL-1β and 10 mg/L NaI for 48 h were detected by flow cytometry. Results The positive expression rate of CD54 in Hashimotos thyroiditis tissues was much more than that of control tissues ( P 〈 0.05 ), and positive areas incresesd as well ( P 〈 0.05 ). The expression of CD54 in the isolated thyroid cells of Hashimotos thyroiditis tissues was more than that of control tissues, but without significant difference. Being interferred by different agents, the isolated cells from Hashimotos thyroiditis tissues expressed more CD54 than that from control tissues ( P 〈 0.05 ). IL-1β of high concentration could stimulate higher CD54 expression in cells from control tissues, and IL-1β even of low concentration had a more stronger effect on the cells from Hashimotos thyroiditis tissues. The combination of NaI and IL-1β increased CD54 expression more significantly. Conclusion It was a secondary response that CD54 expression increases in Hashimotos thyroiditis, and may be related to IL-1β and NaI. CD54 plays an important role in pathogeny of Hashimotos thyroiditis.
