Separation of Zopiclone Enantiomers by HPLC Using β-Cyclodextrin as Chiral Mobile Phase Additive

摘要 目的:建立HPLC手性流动相添加剂法拆分佐匹克隆对映体。方法:选用ZorbaxRx-C18色谱柱(4.6mm×250mm,5μm);流动相为0.12mol/L磷酸二氢钠缓冲液甲醇(7525,V/V),其中含9.0mmol/L的β-环糊精,pH为6.0,流速为0.8ml/min;检测波长305nm;进样量20μl;柱温为20℃。结果:建立了β-环糊精手性流动相添加剂法拆分佐匹克隆对映体的方法。结论:该方法可以达到基线分离,且操作简便,比手性柱法更经济。 Objective: To establish a HPLC method to separate zopiclone enantiomers using chiral mobile phase additives Methods: Separation was performed on Zorbax Rx-C18 column (4.6 mm ×250cm,5μm) with mobile phase composed of 0.12 mol/L of NaH2PO4-Methanol (75:25) mixed 9.0 mmoL/L β-cyclodextrin pH was adjusted to 6.0, the flow rate was 0.8 ml/min and the detection wavelength was set at 305 nm. Results:Using β-cyclodextrin as chiral mobile phase additive, a method was established to separate zopiclone enantiomers. Conclusion: The method is simple and economic with good resolution.
