摘要
目的:观察哮喘豚鼠气道重构模型气道内胰岛素样生长因子-Ⅰ(IGF-Ⅰ)的蛋白表达及电针对其的影响。方法:将48只豚鼠随机分为4组:正常对照组、电针组、地塞米松组、模型组,每组12只。除正常对照组外,其他各组第1天以10%卵清蛋白注射液腹腔注射致敏,于第15天起隔天用1%卵蛋白溶液超声雾化以激发(共6周)进行造模。干预治疗采用电针背六穴(大杼、风门、肺俞,均为双侧),隔天1次,共6周。治疗结束后,采用免疫组织化学法检测与气道重构相关的细胞因子IGF-Ⅰ在各组支气管和肺组织中的蛋白表达。结果:模型组IGF-Ⅰ阳性细胞数明显高于正常对照组,2组比较,差异有非常显著性意义(P <0.01);电针组与地塞米松组IGF-Ⅰ阳性细胞数明显低于模型组,与模型组比较,差异有显著性意义(P<0.05)。结论:哮喘豚鼠气道重构模型气道内IGF-Ⅰ的蛋白表达增加,电针豚鼠背六穴能降低气道重构豚鼠模型支气管和肺组织中IGF-Ⅰ蛋白表达,提示电针豚鼠背六穴可通过下调IGF-Ⅰ蛋白表达,以干预哮喘豚鼠的气道重构。
Objective: To observe the protein expression of insulin growth factor-Ⅰ(IGF-Ⅰ) and influence of electro - acupuncture in air passage of reconstituted model of asthmatic guinea pigs. Methods: 40 guinea pigs were allocated to normal control group (NCG), electro -acupuncture group (EAG), dexameson group (DG) and model group (MG) (each group 12 guinea pigs). Except the NCG, 10% of egg albumin was applied in other groups for intraperitoneal injection in the first day to cause allergy, from the 15th day, 1% of egg albumin solution was applied for ultrasonic nebulization in every 2 days and lasting for 6 weeks to establish models. The electro - acupuncture of Beiliu points (bilateral Dazhu (BL 11), Fengmen (BL 12) and Feilshu (BL 13) points were applied in every 2 days and lasting for 6 weeks as intervention treatment. The end of treatment, the protein expression of air passage reconstituted IGF-Ⅰin bronchi and lung tissue of various groups were determined by immuno-histochemi-cal method. Results: The positive cell count of IGF -Ⅰof MG was marked increased than NCG ( P < 0. 01); the positive cell count of IGF -Ⅰof EAG and DG were marked decreased than MG ( P < 0. 05). Conclusion: The protein expression of IGF -Ⅰof air passage reconstituted models of asthmatic guinea pigs was increased, electro - acupuncture Beliu points in guinea pigs can decrease the protein expression of IGF -Ⅰin bronchi and lung tissue of air passage reconstitution of guinea pigs models. It is indicated that electro - acupuncture Beiliu points of guinea pigs can decrease protein expression of IGF -Ⅰto intervene the reconstitution of air passage of asthmatic guinea pigs.
