摘要
目的探讨胃癌形成中核转录因子κB P65(nuclear factor kappa B P65,NF-κB P65)和尿激酶型纤溶酶原激活物(urokinase-type plasminogen activator,uPA)的表达规律及相关性。方法应用免疫组织化学SABC法检测64例标本中胃癌、癌旁及正常组织RelA和uPA表达情况,采用Pearson等级相关检验研究两者之间相关性。结果RelA在胃癌、癌旁、正常组织表达的阳性率分别为53.13%(34/64)、31.25%(20/64)和7.81%(5/64),三者比较有显著差异(P<0.01);uPA阳性率在上述三者分别为59.38%(38/64)、39.06%(25/64)和12.50%(8/64),亦有显著性差异(P<0.01)。RelA和uPA在低分化腺癌、黏液腺癌、印戒细胞癌中表达增强,与乳头状腺癌、管状腺癌相比有显著性差异(P<0.01,P<0.05)。Pearson相关分析显示RelA和uPA的表达水平在胃癌(P<0.01)、癌旁组织(P<0.01)及正常组织(P<0.01)中均呈显著正相关。结论RelA在人胃癌组织中与邻近组织相比是超表达的,其靶基因uPA也被上调,共同参与了胃癌的形成和发展。
Objective To investigate the expression of nuclear factor kappa B P65 [NF-kB P65 (RelA)] and urokinase-type plasminogen activator (uPA) in gastric carcinogensis and their relationship. Methods Sixty-four sections from gastric cancer patients who had an operation as experimental group, at the same time adjacent noncarcinoma and normal tissue speciments as control groups. The expressions of RelA and uPA are detected immunohistochemistrically by SABC technique. The correlation between RelA and uPA was analized by Pearson analysis. Results The positive rates of RelA and uPA in gastric cancer, adjacent noncarcinoma and normal tissues was 53. 13 % and 59.38 %, 31.25 % and 39.06 %, 7.81 % and 12.50 % respectively. Diffrences were significant(P〈0.01 and P 〈0.05). RelA and uPA overexpressed in poorly differentiated adenocarcinoma,mucoid adenocarcinoma and signet ring cell carcinoma contrasting to polypoid adenocarcinoma and tubular adenocarcinoma ( P 〈0.01 and P 〈0.05). Pearson analysis indicated a positive correlation of RelA and uPA in gastric cancer tissiues( P〈0.01, rp =0. 6840) , adjacent noncarcinoma tissues ( P〈0.01, rp =0. 3368) and normaltissues( P〈0.01, rp =0. 5499). Conclusion The expression of RelA is significantly increased in gastric cancer tissues, compared to adjacent noncarcinoma and normal tissues and its target gene uPA was upregulated,which may contribute to the formation and the progress of gastric cancer.
