摘要
目的探讨在散发性乳腺癌中乳腺癌易感基因BRCA1表达的意义及其与原癌基因C-erbB-2、增殖核抗原Ki67蛋白表达的关系。方法采用免疫组化法对47例散发性乳腺癌组织进行BRCA1及C-erbB-2、Ki67、雌激素受体(ER)、孕激素受体(PR)检测,并分析BRCA1与临床病理指标及C-erbB-2、Ki67的关系。结果BRCA1在散发性乳腺癌中的表达明显下降或缺失,其失表达率为46.8%,表达定位于细胞核及细胞质。BRCA1的表达与C-erbB-2的表达呈负相关(P<0.05),与Ki67的表达无关(P>0.05),与绝经状况、肿块大小、ER、PR状况及淋巴结转移无关(P>0.05)。结论BRCA1的表达下降或缺失与散发性乳腺癌的发生发展有关,BRCA1与C-erbB-2共同检测有望成为预测乳腺癌预后及指导治疗的生物学指标。
Objective To study the expression of BRCA1 gene and the relationship between BRCA1 and C-erbB-2, Ki67 in sporadic breast cancer. Methods Immunohistochemistry method was used to detect the BRCA1, C-erbB-2, Ki67, ER, PR protein in paraffin embedded breast cancer tissue of 47 patients with sporadic breast cancer. Results The expression rate of BRCA1 protein was 46.8% in breast cancer, which was mainly located in the nuclear and cytoplasm of cancer cells. The loss of BRCA1 protein was associated with the expression of C-erbB-2 protein, but did not correlate with Ki67 protein and menopause status, tumor size, ER, PR status and lymph node metastasis. Conclusion Down regulation of BRCA1 protein may be involved in the occurrence and development of breast cancer.
