缺血再灌注后股骨头骺关节软骨的形态学观察

摘要 目的对发育期髋关节缺血再灌注后股骨头骺关节软骨的形态学变化进行观察。方法80只4周龄SD大鼠随机分为缺血再灌注组及对照组,开腹暴露腹主动脉并关闭3h,制作髋关节缺血一再灌注模型,于术后不同时点取股骨头骺关节软骨进行透射电镜观察。结果缺血再灌注组术后光镜观察到关节软骨的退行性改变,透射电镜观察到膜性细胞器的损害及软骨基质成分的变化,表现为结构模糊、线粒体肿胀、内质网扩张、核蛋白体及糖原颗粒减少,软骨基质胶原纤维增粗、间质颗粒减少、钙盐沉积增多。结论发育期髋关节缺血再灌注后股骨头骺关节软骨的损害表现为一定程度的退行性改变,缺血再灌注后超微结构的损害是关节软骨退行性改变的基础,抑制再灌注损伤可能有利于防止发育期关节软骨的退行性改变。 Objective To observe the morphological changes of articular cartilage of femur head epiphyses after ischemia-reperfusion. Methods Four-weeks Sprague-Dawley rats were classified randomly two groups: ischemia-reperfusion group and control group, articular cartilage of femur head epiphyses were examed by light microscope and electron microscopy in different time. Results The hght microscopy showed that chondrocyres degeneration and reduction, as well as fibrosis in matrix of cartilage in the ischemia-reperfusion group. The ischemia-reperfusion group showed that membranes of cell organ and matrix of cartilage have changed by electron microscopy, those change included mitochondria swelling, endocytoplasmic, reticulum enlargement, ribosome and glycogenosome reduction, collagen fibrils thickening, matrix particles reduction and calcium salts deposition increase. Conclusion Femur head epiphyses after ischemia-reperfusion would cause degeneration of articular cartilage, the alteratons of morphological of cartilage tissue are basis of the degeneration of articular cartilage of femur head epiphyses, it have shows that inhibition of ischemia-reperfusion injury to articular cartilage in development stage may be useful for early osteoarthritis.
