^125I-NGF在小鼠体内的血药浓度-时间过程

摘要 本研究涉及应用Idogn法对神经生长因子(NGF)进行^125I-NGF标记,并用同位素示踪法与电泳法相结合的方法研究^125I-NGF在小鼠体内的血药浓度-时间过程。结果显示,静脉注射^125I-NGF在小鼠体内的代谢规律符合二房室开放模型。分布相半衰期为0.13h,消除相半衰期为3.68h,^125I-NGF在小鼠体内分布和消除均较快,清除率为0.125L·h^-1·kg^-1,表观分布容积为0.697L·kg^-1,曲线所匍面积为16.01μg·h^-1·L^-1。 The study concerns in the label of nerve growth factor(NGF) by ^125I with Idogn method. Course of time - ^125I-NGF concentration in plasma of mouse was investigated with the method of sodium dodeeylsulfate-polyaeerylamide gel eleetrophoresis (SDS-PAGE) after iv ^125I-NGF. Result demonstrates the curve of time- ^125I-NGF concentration in plasma after iv fit a two compartment open model. Distribution phase half life is 0.13 h, elimination phase half life is 3.68 h, the mean plasma clearance is 0.125 L·h^-1·kg^-1, apparent volume of distribution is 0.697 L ·kg^-1, and the area under curve is 16.01μg·h^-1·L^-1.
