Iloprost液4℃保存兔胎脑皮层组织的细胞存活率及腺苷磷酸酯的含量变化

摘要 以Iloprost液、生理盐水为保存液,对4℃保存兔胎脑皮层组织进行对比研究,以了解4℃保存的细胞存活率及能量代谢情况。结果表明:①随保存时间延长,细胞存活率逐渐下降,Iloprost液保存的细胞存活率明显高于生理盐水;②三磷酸腺苷(ATP)、能量负荷(EC)随保存时间延长明显下降,在保存2h后接近耗竭,两组间差异不显著(P>0.05)。从以上结果,我们认为Iloprost液的保存效果优于生理盐水,但保存时间仍有限,应考虑在细胞保护剂的基础上加用新陈代谢抑制剂,从而降低代谢速度。 In order to demonstrate the survival rate and energy metabolism of the cells of brain cortex preserved at 4 ℃, a comparative study was undertaken between the rabbits′ embryonic brain cortex preserved with iloprost solution (50 ng/ml) and that preserved with saline at 4 ℃. It was found that: (1) The cell survival rate of the preserved cortex was decreased with the prolongation of the preserving time. The cell survival rate in iloprost solution was significantly higher than that in the saline. (2) Adenosine triphosphate (ATP) and energy charge (EC) level was obviously decreased when the preserving time was prolonged and tended to be exhausted on the second hour after preservation. The difference between the two groups was not significant statistically ( P >0.05). Through the analysis of these findings, it was concluded that iloprost solution was better than saline in preserving the rabbits′ embryonic brain cortex but its preserving time was still limited. It is suggested that some metabolism inhibitors be added to the cell protective agents to decrease the metabolic speed and improve the preserving effects at 4 ℃.
