摘要
目的选育核糖霉素高产菌株,提高其生产发酵水平。方法以核糖苷链霉菌03-8为出发菌株,经紫外线和氯化锂复合处理后,再采用微波诱变,筛选自身产物抗性解除反馈调节突变株,同时结合单菌落自然分离纯化法进行选育。结果获得了稳定高产菌株563-44,比出发菌株产量提高201.1%,其生产能力在5吨发酵罐试验中得到验证。结论变异株563-44确实是1株高产的工业生产菌种。
Objective To increase the fermentation yield, a high producing strain of ribostamycin was selected. Methods The parent strain streptomyces ribosidificus 03-8 was mutated with the combined treatment by UV, LiCl and microwave. Mutant strain was selected by self-product resistivity to dismantle feedback regulation, and the method of single colony isolation was also used. Results A high producing strain 563-44 was obtained. The yield of the mutant in the shaking flask was about double higher than the parent strain. Trail-production in a 5 tonfermenter proved its productivity. Conclusion The strain 563-44 is indeed a high producing strain for industrial production.
