人成釉细胞瘤中蛋白激酶C-α表达与端粒酶活性的关系 被引量:6

摘要 目的:研究蛋白激酶C-α(PKCα)在人成釉细胞瘤(AB)中的表达及与端粒酶逆转录酶(hTERT)的关系,探讨其临床生物学意义。方法:用原位杂交和免疫组化SP法分别检测了AB中hTERT mRNA及PKCα蛋白的表达。同时选取牙源性角化囊肿(OKC)16例,正常口腔黏膜7例做对比研究。结果:hTERT mRNA在AB中阳性率为94.4%(51/54),OKC中为87.5%(14/16)及1/7正常口腔黏膜有hTERT mRNA表达,组间差异有显著性(P<0.001)。伴随AB的复发与恶变hTERT的表达逐渐增高。PKCα在AB中阳性表达为85.1%(46/54),OKC中为56.2%(9/16),4/7正常口腔黏膜有PKCα阳性,组间差异有显著性(P<0.01),伴随AB的复发与恶变PKCα阳性表达也随之增高(P<0.05)。hTERT阳性表达与PKCα阳性表达之间经Kendall相关分析rk=1.000,P=0.005呈高度正相关。结论:hTERT与AB的发生发展及临床生物学行为相关,端粒酶活性的释放激活与PKCα的高表达可能正相关。 Objective: To investigate the expression of protein kinase C-α (PKC-α) in human ameloblastoma (AB) and its relationship with human telomerase reverse transcriptase (hTERT). Methods: The expression of hTERT mRNA was detected by in situ hybridization and PKC-α protein by immunohistochemistry in 54 cases of AB, 7 cases of normal oral mucosa(NOM) and 16 cases of odontogenic keratocyst ( OKC ). Results : The positive rate ofhTERTmRNAinAB, OKC and NOM was 94.4%(51/54), 87.5%(14/16) and 14.3%(1/7)(P〈 0.001 ) respectively. The positive rate of PKC-α in AB, OKC and NOM was 85. 1% (46/54), 56.2% (9/16) and 57.1% (4/7) ( P 〈0.01 ) respectively. In recurrent and malignant AB both hTERT and PKC-α expressions were higher than those in primary AB(P 〈0.05 ). hTERT and PKC-α showed positive relation proved by Kendall correlation analyze (rk= 1. 000 ,P = 0. 005 ). Conclusion:hTERT plays an important role in genesis and development of AB. Telomerase activity is positively correlatied with PKC-α expression in AB.
