HIF-1α、GLUT-1在骨肉瘤中的表达及其与血管生成的关系研究 被引量:4

摘要 目的探讨缺氧诱导因子-1α(HIF-1α)与葡萄糖转运蛋白-1(GLUT-1)在骨肉瘤组织中的表达,并分析两者的相关性,同时研究两者在骨肉瘤血管形成中所起的作用。方法采用免疫组织化学S-P法,检测64例骨肉瘤和10例瘤旁正常骨组织、6例骨纤维结构不良HIF-1α与GLUT-1蛋白的表达,同时用CD34标记血管内皮细胞,计算微血管密度(MVD)。结果64例骨肉瘤HIF-1α蛋白阳性表达率为64.1%(41/64),与对照组(12.5%,2/16)比较有非常显著性差异(P<0.01),GLUT-1阳性表达率为60.9%(39/64),亦显著高于对照组(12.5%,2/16),(P<0.01);HIF-1α与GLUT-1表达一致符合率为57.8%(37/64),两者表达显著相关(P<0.05),且与瘤组织分期显著相关(P<0.05)。MVD平均值为21.43±8.56,HIF-1α阳性的骨肉瘤组织MVD值(32.43±8.32)明显高于阴性者(16.67±6.56),两者比较有非常显著性差异(P<0.01)。结论骨肉瘤组织存在HIF-1α与GLUT-1蛋白的过表达,两者在骨肉瘤的发生、发展过程中可能具有协同作用,HIF-1α对骨肉瘤新生血管生成可能有重要作用。 Objective To study the relationship of the expression of HIF-laand GLUT-1 with angiogenesis in osteosarcoma, and to analyizc the correlation of both. Methods HIF-1αand GLUT-1 protein expression were dctected by immunohistochcmical methods in 64 cases of osteosarcorna and 10 cases of normal bone tissue and 6 cases of osteofibrous dysplasia, CD34 marking vascular endothelialccll was used to measurc thc micro-vascular dcnsity( MVD ). Results The poisitive cxpression rate of HIF-1α and GLUT-1 were 64.1% (41/64) and 60.9% (39/64) in 64 cases of osteosarcoma ,showing a significantly difference in thc expression of HIF-1α and GLUT-1 betwecn osteosarcoma and control group(both are 12.5 % );The coincidcnce rate of the exprcssion of HIF-1α and GLUT-1 was 57.8% (37/64) ,showing a close relation between them,and the expression of HIF-1α and GLUT-1 were significantly correlated with stage of the tumor. The medium MVD was 21.43 ± 8.56, much higher in tumor with possitive expression of HIF-1α(32.43 ±8.32 ) than in tumor with negative expression of it (16.67 ± 6.56), there was a significantly difference between them. Conclusion There have overexpreasion of HIF-1α and GLUT-1 in ostcosarcoma, they may have a synergistic effect in the genesis and development of osteesarcoma,HIF-1α may play an important role in angiogcncsis of ostcosarcoma.
