摘要
采用不连续聚丙烯酰胺垂直板电泳技术,分析了花鱼鳃、眼睛、肌肉3种组织中6种同工酶差异表达,并对部分同工酶位点表达及酶谱表型进行了初步分析.实验结果显示:SDH在3种组织中均未见表达;ADH仅在肌肉组织和眼睛中出现较弱的1条酶带;其它4种酶在3种组织均有较好地表达,其中EST、SOD在组织间表达不存在明显差异;而LDH、MDH同工酶在酶带数及酶活性方面有明显的组织特异性,这可能是受不同基因位点或同一基因位点不同等位基因控制的结果.在6种同工酶中共检出7个基因座位.
Six isozymes, extracted from three tissues of Hemibarbus maculates Bleeker respectively, were analysed with vertical slab polyacrylamide gel electrophoresis. The isozymic phenotypes, tissue distributions, relative activities and mobile characteristics of the six isozymes were studied, and the possible relationships between the isozyme pattern and gene expression were discussed. The results showed: SDH had no gene expression ; ADH was only found one weak enzymes band in foot muscle and eye; four other kinds of isozymes had extreme expression; EST and SOD had no tissue distributions, while LDH and MDH had remarkable tissue distributions, which may be controlled by diferent gene loci or different alleles in the same gene locus. Altogether 7 gene loci were detected in 6 kinds of isozymes.
