摘要
目的:研究基质金属蛋白酶(MMPs)及其抑制因子(TIMPs)在前列腺癌转移侵袭中的作用。方法:采用免疫组织化学、免疫荧光双重染色、形态定量分析方法对5例正常前列腺组织和34例前列腺癌组织MMPs及TIMPs进行定性、定位、定量检测。结果:免疫组织化学染色显示,随着前列腺癌恶性程度的提高,MMP7和MMP9的表达均增强(P<0·001)。TIMP1和TIMP2前列腺癌组织的GleasonGrade分级间表达差异均无显著性(P>0·05)。免疫荧光染色显示,MMP13前列腺癌细胞和正常前列腺腺细胞异的胞膜上均有表达。MMP7和LN可共同表达于前列腺癌细胞上、细胞基质中的基底膜、血管平滑肌和神经纤维上。MMP9和Ⅳ型胶原主要表达于细胞外基质中的基底膜上。结论:MMP7和MMP9可作为前列腺癌侵袭和转移的判定指标。MMP13和肿瘤恶性程度无关联性。前列腺癌的不同进展期TIMP1和TIMP2的变化与MMPs不同步。
Objective To study the biological effects of matrix metalloproteinases (MMPs) and tissue inhibitor of metalloproteinases (TIMPs) in invasion and metastasis of prostate carcinoma. Methods Immunohistochemistry and double- immunofluorescence techniques were used to detect the expressions of MMPs and TIMPs in 5 normal prostate and 34 prostate carcinoma tissues. Results Immunohistochemistry showed that the expressions of MMP7 and MMP9 increased with malignant degree of prostate carcinoma (P〈0. 001). TIMP1 and TIMP2 expressions had not significant difference between normal prostate and prostate carcinoma (P〉0.05). Immunofluorescence staining showed that MMP13 expressed in membrane of normal prostate cells and cancer cells. Double immunofluorescence staining showed that MMP7 and LN both expressed in cancer cells, basement membrane, smooth muscle cells and nerve fiber. MMP9 and collagen IV both expressed in basement membrane. Conclusion MMP7 and MMP9 can be used as markers for invasion and metastasis of prostate carcinoma . MMP13 has nothing with malignant degree of tumor. In different grades of prostate carcinoma , TIMPs and MMPs have different changes.
