摘要
利用3-’RACE的方法分别从异源四倍体鲫鲤、三倍体湘云鲫、二倍体红鲫的卵巢中克隆了cdc2基因cDNA的部分序列.选取这3种鱼cdc2 cDNA中一段长为157 bp的保守片段设计特异引物,通过Real-time PCR检测cdc2 mRNA在这3种不同倍性鱼卵巢中的表达水平,结果表明cdc2转录量随着倍性的递增而相应升高.
The partial sequences of cdc2 cDNA for different ploidy level cyprinid fish (allotetraploid hybrids, allotriploid crucian carp and red crucian carp ) were cloned by 3'-RACE ( Rapid Amplification of cDNA Ends) method and the amplified fragments were sequenced. Real-time PCR was used to detect the cdc2 mRNA in the ovaries using specific primer sets synthesized on the conserved region with the length of 157bp of cdc2 cDNA among these different ploidy level cyprinid fish. The result indicated that the transcriptional level of cdc2 mRNA was up-regulated as the ploidy increases.
