肾康丸对高糖培养的大鼠系膜细胞TGF-β_1及FN的影响 被引量:1

摘要 目的:观察肾康丸对高糖培养大鼠系膜细胞(MC)分泌转化生长因子(TGF-β1,及纤连蛋白(FN)分泌mRNA表达的影响,探讨其防治DN的分子机制。方法:体外培养大鼠MC,将细胞分为6组:低糖组、高糖组、正常血清组、开搏通组、肾康丸高、低剂量组。培养72 h后,采用双抗夹心酶联免疫吸附(ELISA)法检测高糖对MC分泌TGF-β1和FN的影响;采用逆转录-多聚酶链反应(RT-PCR)法检测高糖对TGF-β1和FN mRNA的表达的影响;同时研究药物血清对上述指标的影响。结果:高糖可以促进MC分泌TGF-β1和FN,并提高MC中TGF-β1和FN mR-NA的表达,肾康丸可以明显抑制上述作用。结论:肾康丸可以通过TGF-β1途径抑制MC基质分泌。 Objective: To investigate the effect of Shenkangwan on secretion of transforming growth factor-β1 (TGF-β1 ) and Fibronectin( FN )and their mRNA expression in rat mesangial cells( MC )cultured by high glucose and explore its molecule mechanism on treating diabetic nephropathy(DN). Methods: Rat MC were cultured in vivo and divided into six groups: low glucose group, hight glucose group, normal rat serum group, Capoten tablets group, Shenkangwan high dose group and low dose group. Seventy two hours later, the secretion of TGF-β1 and FN in MC were detected by enzyme linked immunosorbent assay(ELISA). The expression of TGF-β1 and FN mRNA were determined by semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR). Results: High gluscose could induce secretion of TGF-β1 and FN in MC and increase expression of TGF-β1 and FN mRNA. While Shenkangwan could repress these effects. Conclusion: Shenkangwan can suppress the secretion of the extracellular matrix in MC via TGF-β1 signal way.
