摘要 目的:探讨中药复方制剂DSF对于心肌缺血大鼠血液及组织中NO系统的影响。方法:将40只大鼠随机分为对照组,模型组,DSF组和丹参片组。用大鼠冠状动脉左前降支结扎的手术方法制备急性心肌缺血缺氧模型,以试剂盒法测试血清及心肌组织中的NO、NOS、iNOS、cNOS的含量;用免疫组化的方法测定DSF对心肌组织中的cNOS mRNA表达的影响。结果:与模型组比较,DSF组血清NO及缺血区心肌组织NO含量均有明显升高(P<0.05或P<0.01),DSF组血清及缺血区心肌组织NO,cNOS含量有明显升高,iNOS含量没有明显差异但其与NOS之比明显降低,说明其相对含量比模型对照组有所下降,并且缺血区心肌组织cNOS mRNA表达对与模型对照组比较也有显著增高(P<0.05)。结论:DSF能明显提高NO水平,升高cNOS含量,降低iNOS/NOS比例,其抗心肌缺血作用机制可能是通过提高机体NO含量及NOS活性,提高cNOS mRNA的表达来实现的。同时DSF也可能通过其降低iNOS作用对心肌起保护作用。 Objective: To observe the effect of DSF on NO System in experimental myocardial ischemia model by ligating the cornary descending branch. Methods: Wistar mice were divided into control group, model group, Danshen group and DSF group to observe the effect of DSF on variations of the NO system. Results: Compared with model group, content of NO, NOS and cNOS in serum and muscle in DSF group were significantly increased (P 〈0. 05 或 P 〈0. 01 ). But there was no statisical difference on the content of iNOS ( P 〉 0. 05 ) compared with model group. The expression of cNOS mRNA was increased ( P 〈0. 05 ). Conclusion : The mechanism of DSF in treating coronary is realized by improving the content of NO, the activity of NOS and expression of cNOS mRNA in myocardial tissue.
