Whether plantS contain protein kinase C (PKC) is still uncertain and isan interesting question in the field ofsignal transduction in plant. In order toobtain more evidence for it, we employed immunofluorescence and 32P-labeling which had been used to determinethe activity of PKC in animal cells, theetiolated seedlings of gram (Phaseolus radiatus L. )and the cotyledon callus of kiwi fruit (Actinida deliciosa Liang. ) wereused as materials. The following resultshave been obtained:1. After the protoplasts isolatedfrom green gram epicotyl and kiwi fruitcallus were incubated with the antibodyto mouse brain PKC, high intensity offluorescence was found in regions ofcell membrane and nuclei under fluorescence microscope (Figs. 1, 2 ). Thisshows that PKC exists in plant cells.2. Ca2+ and phosphatidylserineactivated PKC were measured as the 3Tradioactivity level in histone phosphorylation. The activity of PKC was 17. 00Bq mg-1 prot min-1. However, the activity was decreased by 25. 8% bytreatment with H7, an inhibitor ofPKC, at a level of 1 umol/L, and31. 7% of the total activity of PKC remained when H7 was at a concentrationas high as 100μmol/L (Fig. 4). Thisresult suggests that PKC in plant cellsmight be similar to that in animal cells.
