摘要
利用无核白和黑王等12个品种的无病毒葡萄试管苗叶片为外植体,在NN69培养基(N itsch and N itsch 1969)试验了不同浓度的IBA和BA对其植株再生的影响。其中无核白、黑王、红地球和红宝石4个品种获得了稳定高频率的再生植株。以GUS基因为报告基因,通过根癌农杆菌介导,建立了葡萄离体叶片外源基因的遗传转化体系,而后将葡萄扇叶病毒外壳蛋白(GFLV-CP)基因转入葡萄叶片,通过卡那霉素筛选和PCR检测证明获得了转葡萄扇叶病毒外壳蛋白(GFLV-CP)基因的无核白葡萄植株。
On the basis of high and stable plant regeneration from the leaves of " Tomson Seedless", "Heiwang", "Red Globe" and "Ruby Seedless" on the medium of Nitsch and Nitsch ( 1969), the transformation system of grape was established with GUS gene via Agrobacterium tumefaciens. The transgenic plant of "Tomson Seedless" was obtained with GFLV-CP gene by the selection of kanamycin and Conformed by PCR detection.
