百合种质资源限制生长法保存研究 被引量:37

摘要 对切花百合(Lilium L.)品种‘Siberia’(西伯利亚)试管苗限制生长保存方法及保存后恢复生长的再生植株的遗传稳定性进行了研究。结果表明,常温(20±1)℃、光照强度1500lx、光照时间12h/d的条件下,在MS或者1/2MS培养基中添加1.0~3.0mg/L的脱落酸(ABA)能够有效延长百合试管苗的继代间隔时间。离体保存百合试管苗9个月时存活率均在80%以上。在添加50~90g/L高浓度蔗糖的培养基中能够保存百合试管苗11个月以上,保存过程中植株生长缓慢,基部有紫色鳞茎生成。而添加10~50g/L甘露醇或10~40mg/L矮壮素(CCC)对百合试管苗生长没有起到明显的限制作用。各种限制生长法保存的试管苗转移到恢复培养基上培养1个月左右均能长成正常的植株,其形态特征以及可溶性蛋白质、过氧化物酶同工酶和酯酶同工酶图谱与对照相比没有明显差异。 Studies on conservation of in vitro shoots of lily (Lilium L. ) by restricting growth method and genetic stability of recovered plantlets were carried out. Results showed that 80% of in vitro shoots could be conserved in the medium supplemented with 1.0 -3.0 mg/L ABA for more than 9 months at normal temperature of (20 ± 1 )℃, 1 500 lx and 12 h/d photoperiod. After cultured in recovering medium for 1 month, all conserved in vitro shoots turned to normal plantlets and showed no obvious difference in morphology, soluble protein, isoenzynic zymogram of esterase and peroxidase compared with the control.
