摘要
cDNA文库是分离、筛选基因的重要平台。利用SMART^TM cDNA library construction技术,成功构建了金钗石斛花芽cDNA表达文库。结果表明该文库的克隆数为3.02×10^5pfu,插入片段的大小在0.5~2.5kb范围,插入片段的重组率接近100%,说明所构建的cDNA文库质量较好。该文库的构建为克隆与金钗石斛花芽分化相关功能基因奠定了基础。
A cDNA expression library from the /lower bud of Dendrobium nobile was constructed into pcDNA3.0 eukaryotical expression plasmid used SMARTTM cDNA library construction protocol. The result showed that the cDNA expression library contained 3.02 × 10^5 pfu, the size of the inserts ranges from 0. 5 kb to 2.5 kb, and the percentage of positive clones was almost 100%. All of the above mentioned have answered for the general requirements of a cDNA library. This provides a base for cloning of important genes related to flower bud differentiation in Dendrobium nobile.
