摘要 目的:探讨趋化因子受体在前列腺癌组织中的表达及其意义。方法:采用酶标记免疫组织化学方法检测45例前列腺癌及10例前列腺增生组织中趋化因子受体CCR1、CCR3、CXCR4和CCR5的表达情况,应用ELISA双抗体夹心方法检测相应患者血清中趋化因子SDF-1的含量,并应用免疫发光方法检测相应患者血清中前列腺特异性抗原PSA的含量。结果:在前列腺癌组织上检测到趋化因子受体CXCR4的表达(表达率55.5%),PSA>20 ng/ml与PSA<20 ng/ml的前列腺癌组织上CXCR4的表达率分别为61.2%和42.8%,而前列腺增生组织中未发现这4种趋化因子受体的表达;与前列腺增生患者相比,在前列腺癌患者血清中SDF-1含量明显升高[(567.9±90.73)vs(169.1±46.01)pg/ml,P<0.01]。结论:在前列腺癌组织上发现有趋化因子受体CXCR4的表达,其表达可能在前列腺癌的发生、发展和转移中起重要作用。 Objective:To detect the expression of chemokine receptor CXCR4 in human prostatic carcinoma and explore the role of CXCR4 in prostate cancer. Methods: Forty-five prostatic carcinoma specimens and 10 prostate hyperplasia specimens were immunohistochemically stained for CCR1,CCR3 ,CXCR4 and CCR5 expression. SDF-1 concentrations were measured by ELISA and PSA concentrations were measured by immune luminescence in the sera of patients. Results: CXCR4 expression was detected in prostatic carcinoma tissues (the positive rate was 55.5%) but not in prostate hyperplasia. The positive rate of CXCR4 was 61.2% when PSA concentrations 〉20 ng/ml and was 42.8% when PSA concentrations 〈20 ng/ml. SDF-1 concentrations in the patients with prostate carcinoma increased significantly compared to patients with prostate hyperplasia(P〈0.01). Conclusion.. CXCR4 is expressed in human prostatic carcinoma, and it may play an important role in the development,advancement and metastasis of human prostatic carcinoma.
