维生素E对兔假体包膜形成的抑制作用 被引量:14

摘要 目的:探讨维生素E干预假体包膜形成的作用。方法:12只雌性成年新西兰兔,随机分为2组,每组6只,每只兔于肉膜下置入15ml光面硅胶假体4枚。对照组饲喂普通饲料,维生素E组术后饲喂添加大剂量维生素E的饲料。3个月后取假体周围包膜检测包膜张力、包膜重量、包膜厚度、包膜组织中胶原含量、α-平滑肌肌动蛋白免疫组化染色阳性细胞数量并进行包膜组织学检查。结果:对照组包膜张力、包膜重量、包膜组织中胶原含量、α-平滑肌肌动蛋白免疫组化染色阳性细胞数量均高于实验组(P<0.01),2组包膜厚度无差异(P>0.05);2组包膜的显微结构形态基本一致,内层较平滑,中层是成层排列的纤维结构,外层由疏松结缔组织构成;对照组胶原纤维致密,中层较实验组厚;2组包膜中均可见到典型的肌成纤维细胞,实验组可见肌成纤维细胞线粒体肿胀、内质网扩张。结论:大剂量维生素E有干预假体包膜形成的作用, Objective To explore the effects of vitamin E on inhibition of periprosthetic capsular formation. Methods 12 female New Zealand white rabbits were randomly divided into two groups with 6 animals in each group. Four smooth silicone implants (15ml each) were implanted beneath the panniculus carnosus muscle of each rabbit. The experimental animals were fed with forage with megavitamin E for three months. All animals were sacrificed 3 months after operation. Capsular tensility, wet weight, collagen content and capsular histological examinations were carried out. In addition, the expression of α-smooth muscle actin (α-SMA) were examined by immunohistochemistry. Results Capsular tensility and capsular wet weight and capsular collagen content in the control group were significantly higher than that in the Vitamin E group (P〈0.01). α-SMA positive cell numbers in control group was obviously higher than that in the vitamin E group (P〈0.01). Capsular thickness were no statistical difference between two groups (P〉0.05).The capsular microstructure was similar in two groups. The inner layer of capsule was smooth. The middle layer was lamellated fibrous tissue. The outer layer was loose connective tissue. Collagen fibres in control group was dense, the middle layer was thicker than that in the vitamin E group. Myofibroblasts with swollen mitochondria were found in the Vitamin E group. Conclusion This study verify the efficiency of high dose of vitamin E in inhibiting the capsular formation.
